Izuko the First was reincarnated, same like Izuko II, meanwhile Izuko III's soul was destroyed because she broke the Gate's rule. Meanwhile Izuko IV is still unknown, maybe this sequel⁴'s Izuko is still Izuko IV.
IIRC, Izuko can choose to stay being Izuko as long as she want or choose to reborn when the new Izuko is taking over the duty as the Guardian of the Gate <= That's what happened to Izuko the First and Izuko II, Izuko the First offered a murder's victim soul to stay at the Gate and become the new Izuko, and then Izuko the First reborn.
Tl;dr, Izuko allowed to offers her duty as the Guardian of The Gate to another soul who come to the Gate and Izuko can't die unless she break the Gate's rules which makes her soul destroyed by that Toad (or by the Gods).
